Downspout questions for my new rain barrel

About to install a new rain barrel at my house. My current gutters run the entire length of my house on each side (envision a ranch style house-rectange!) with the downspouts running down near my garage. However, I would like to locate the barrel on the opposite end of the house since that would put it closer to my garden... Would I run into issues by adding a downspout on the opposite end of the house for the barrel? Seems like I might need to block off the current downspout to get enough water. (don't know how I would do that either) I know that most gutters run downhill, so I am thinking this may not work out quite as easily as I want.. Any other advice? I would just like to be able to hook a hose to the barrel and am worried I might run into trouble with a 100' of hose between the barrel and garden.
