Killing grass in autumn

A few years ago, we raised three beds in the middle of our lawn. Two of them were quite high and the grass couldn't grow through. In the third one, however, even though we used a lot of magazine to stop the grass from popping up, after some years, the grass came through. I tried to pull but after two days could only get rid of some of it and it seems impossible to get rid of the roots that are lining the bottom. I intend to dig up the flowers, let the grass grow and and kill it using Roundup. The bed is pretty big. Is it possible to do this in the fall or do I have to wait for spring?

Any other suggestions to get rid of the grass will be also appreciated.
